Investigation of Antibacterial Effect and Expression of Alkaline Protease Gene in Pseudomonas Aeruginosa (PAO1) Using Copper Oxide, Copper Complex and Satureja Khuzistanica: RT-PCR Method

Abstract: Background and objective: In the genus Pseudomondaceae, exists Pseudomonas aeruginosa which is a gram-negative bacterium. It is an opportunistic human pathogen that frequently colonizes patients who are immunocompromised, such as those with cystic fibrosis, cancer or AIDS. Pseudomonas aeruginosa also has inherent resistance to several different chemotherapeutic agents and antibiotics. This study intended to evaluate the inhibition effect of copper oxide nanoparticle (CuO), copper nano complex (Cu complex) and the Satureja Khuzistanica essence on the gene of alkaline protease in P. aeruginosa (PAO1) by RT-PCR method. Method and material: Satureja Khuzistanica essence was synthesized by Clevenger apparatus. Nanoparticular size and morphology were characterized by the use of SEM. The antimicrobial activity of these substances was calculated from the process of disk diffusion and microbroth dilution based on CLSI norms. After determination of the Minimum Inhibitory Concentration (MIC) and Zone of Inhibition (ZOI), these data were analyzed using SPSS 21 software, through descriptive and inferential statistics at a 95% confidence level. In this study, ethical considerations were respected. Results: Zone of inhibition (ZOI) and Minimum inhibitory concentration (MIC) of Satureja Khuzistanica essence, CuO and Cu complex for PAO1 were found to be 19, 15 and 9 mm and 4, 12.5, and 25µg/ml respectively. Furthermore, the MIC synergism of Satureja Khuzistanica essence and CuO was 2 and 3.125µg/ml and Satureja Khuzistanica essence and Cu complex was 2 and 6.25µg/ml respectively. The ZOI synergism of Satureja Khuzistanica essence and CuO was 23mm and Satureja Khuzistanica essence and Cu complex were reported 20mm. Conclusion: According to the results obtained from the present study, a significant antibacterial activity was seen by Satureja Khuzistanica essence. By examining the synergism effect of CuO, Satureja khuzistanica essence, and Cu complex essence simultaneously, the further antimicrobial effect than was observed when each were used alone.
