A New Paradigm Shift in Formulation Development by Smart Drug Delivery System

Abstract:

Development of new formulation for specific response is not an easy task. However, with best efforts over last few decades the technology of drug delivery improved significantly. The continuous efforts of researches added many new chemical entities (NCE) and their respective more improved formulations. If we go through the records, dozens of new potential molecules added in this cascade every year. These new molecules with proven therapeutic effectiveness are approved by various drug approval authorities of the world. On the other hand, some of them were banned due to toxicity and side effects. With some modifications in drug delivery system and advancement in technology, the pharmaceutical researchers are now in a position to develop formulations with more improved therapeutic window and lesser harmful effects compared with conventional drug delivery system which includes tablets, capsules, intravenous injections and other immediate release dosage forms. These conventional dosage forms raise the concentration of drug in plasma quickly and readily declines requiring a second dose to maintain therapeutic level. Because of these fluctuations the level of drug exceeds the maximum effective concentration (MEC) and attain its toxic levels. This is well evident that in such cases drug may also harm healthy cells e.g. chemotherapeutic drugs administered for cancer. With increase in our knowledge in the field of drug delivery system, some smart polymeric material having potential of controlling release of drug using suitable stimulus are now known to us. These polymers are usually polysaccharides having rigid backbone to carry load of drug and capable of releasing drug at suitable sites. Some additional advantages are biodegradability, noncytotoxicity and biocompatibility. The presented review in smart drug delivery system is particularly useful for the researchers having interest in solving problems discussed in preceding lines in the abstract related to conventional dosage forms.
