Optimization Studies on Extracellular Lipase Production from Penicillium citrinum.

Abstract:
|
In the present study, cultural conditions were optimized for maximum enhancement of lipase activity of a novel strain of Penicillium citrinum. The optimum temperature and pH was found to be 40°C and 6.5 respectively. The maximum lipase activity (1.053 ±0.32 IUmL-1) was obtained in the medium containing mixture of fructose (0.5 gm) + Lin seed oil (5.0%w/v) as lipid carbon source and peptone (0.7gm) as nitrogen source. The highest hydrolysis rate were obtained with p-NPP (p-nitro phenyl Palmitate (C16:0) (99.07 ± 1.58) followed by P-NPL (p-nitro phenyl liminate) (C12:0) (96.41 ± 0.92) indicating the enzyme preference for the long chain acyl derivative compounds. The best results were recorded with Ca2+, Mg2+ and Mn2+. But maximum lipase production was achieved with the culture supplemented with Mn2+ (0.147±0.004 IUmL-1) and next with Ca2+ (0.141 ±0.004 IUmL-1). The optimum concentration of Mn2+ was found to be 2mM, at which the lipase activity was recorded, maximum (0.134 ± 0.003 IU ml_1). The lipase showed the optimization of conditions led to 8-fold increase of lipase activity as compared to the initial enzyme activity on basal medium.
