Novel method for simultaneous determination of spiramycin and metronidazole in pharmaceutical dosage form by UV-visible absorption spectrophotometry

A simple, accurate, and rapid UV spectrophotometric method for simultaneous determination of spiramycin SPY and metronidazole MET in binary mixture has been developed.
Two calibration curves were carried out; the first was elaborated to determine MET in the mixture at 320 nm and the second was used to determine the concentration ratio (CR) in the mixture at 230nm. This last was plotted by determination of the absorbance ratio (AR) against the concentration ratio CR= [SPY]/ [MET]. Finally the concentration of the SPY was deduced [SPY] = CRexp × [MET]. The developed method has been validated statistically according to ICH Q2 guidelines and has given satisfactory results. The results of analysis formulation given as percentage of label claim were found to be 99.71% and 99,38 % for SPY and MET respectively for AR1, 99.89 for SPY and 99, 38 for MET for AR2. The suggested method could be used in the routine quality control analysis of pharmaceutical formulation.
