Abstract:

The prime objective of this work is to develop controlled release formulation of glimepiride based on osmosis principle.
The osmotic system comprises of glimepiride, Nacl as osmogent chitosan as release controlling polymer and evaluation was
carried out by in-vitro release studies, burst strength test, IR analysis and stability studies. The concentration of pore forming
agent, effect of osmotic pressure and effect of pH on drug release was also investigated. It was found that a non-zero order
release was observed for 24 h period and the drug release varied with osmotic pressure which linearly relates with the concentration
of pore forming agent. It was also noticed that pore forming agent have direct effect on drug release due to its water
absorption capacity. Chitosan coated on the system was found to control the release rate due to its swelling nature in the dissolution
medium. IR analysis proved the compatibility between drug and polymer. Stability studies indicated the stability of
systems. It was concluded that by using osmotic principle and a suitable natural polymer like chitosan, controlled release drug
delivery systems of other antidiabetic drugs can be developed.
