Abstract:

The purpose of present study was to develop transdermal patch of Diclofenac Diethylamine (DDEA) for its prolonged therapeutic usage. Transdermal patch was optimized by using Box-Behnken design by taking three factors, at three levels. Design expert software is used to find the effect of their independent variables (X1= Polymer concentration (mg), X2= Polyethylene glycol 400 % v/w (PEG-400), X3= Permeation Enhancer (DMSO) % v/w on dependent variables (Y1= in vitro Q24 cumulative amount of drug permeated, mg (CADP In-vitro), Y2=Flux, µg/cm2/hr. Y3= Tensile strength, (MPa). Mathematical equation was generated for Y1Y2Y3 Contour Plots; 3D plots are generated to find the effect of independent variables on dependent variables. Optimized formulation was selected by overlay plots of Response surface methodology. The optimized formulation showed a flux of 376.19 µg/cm2/hr, which could meet target flux.
