Spectrophotometric Determination of Olanzapine by Charge-Transfer Complex Formation in its Pure Form and Pharmaceutical Formulations

Abstract: This study involved rapid, delicate and simple spectrophotometric procedure to determine the drug Olanzapine (Ola.) in its pure form and pharmacological formulations. This reaction was built on Charge-Transfer complex reaction between the basic nitrogen (The more basic secondary amine) of the drug as n-donor with ?-acceptor of the -tetrachloro-1,4-benzoquinone- (Chloranil) (Chl.). The bluish-green colored formed was quantitatively measured at 604 nm. The colored complex obeyed Beer’s law in the concentration range of (1-35) µg.ml . The molar absorptivity value obtained from Beer’s data was found to be 18215 L.mol -1 , Sandell’s sensitivity value was calculated to be 0.0172 µg.cm -1 -1 .cm -2 , while LOD and LOQ were found to be 0.0736 and 0.2229 µg.ml -1 , respectively. The stoichiometry of the complex established between the drug and (Chl.) was 1:1 as estimated by Job’s method of continuous variation and mole ratio method. The association constant (KCT) of the Charge-Transfer complex formative was evaluated using Benesi–Hildebrand equation and was found to be 400 L.mol . The method was successfully applied for the analysis of pharmaceutical formulations.
