Anti-elastase of Leaves and Stem Bark Extract of Garcinia daedalanthera Pierre

Abstract: Purpose: Garcinia daedalanthera Pierre is one of the Clusiaceae family and a native plant from Sulawesi Island, in Indonesia. Elastase enzyme can reduce the elasticity of the skin, causing aging. The study aims to determine the inhibitory activity of the elastase enzyme from G. daedalanthera Pierre. Methodology: The extract was obtained by maceration successively method. The anti-elastase assay using the spectrophotometric method at 405 nm. The enzyme for the anti-elastase assay used porcine pancreatic elastase (PPE), and the substrate used N-Succ-(Ala)-3-p-nitroanilide (SANA). The phytochemical screening for the highest anti-elastase inhibition by qualitatively test. Results: The elastase inhibitory test results of 100 ppm (triplicate) showed that the leaves methanol extract was 23.38 ± 8.38 %, the leaves ethyl acetate extract was 28.91 ± 15.19 %, the leaves hexane extract was 8.14 ± 11.20 %, the stem bark methanol extract was 24.41 ± 5.32 %,and the ethyl acetate extract of stem bark was 43.96 ± 12.53 %. The result of phytochemical screening of the ethyl acetate extract of the G. daedalanthera Pierre stem bark showed that the extract contained flavonoids, alkaloids, tannins, glycoside, and anthraquinones. Applications/Originality/Value: The ethyl acetate extract of G. daedalanthera Pierre stem bark has the highest activity as anti-elastase.
