Abstract:

In past decade cancer therapy researchgavea promissing forward and many facts suggest thepotenteffects of enzymes
depleting amino acids in decreasing the risk of various cancers. In this study L-glutaminase as one of the medically
important enzymes purified from Bacillus subtilisusing80% ammonium sulfate precipitation, then DEAE- celluloseion
exchange chromatography and Sephadex G-200 for gel filtration chromatography. Specific activity of the purified
enzyme by the final purification step was reached to 600 U/mg with 48.8 folds of purification and 45% total
recovery.Enzyme molecular weight of about 48 kDadetermined thought sodium dodecyl sulphate
polyacrylamide.DPPH and ABTS assays in vitro antioxidanttogether withantitumor effects of L-glutaminase against
MCF-7 cell lines by MTT assay indicated an important role of this enzyme in breast cancer.
