ANTI HYPERGLYCEMIC EFFECT OF OCIMUM SANCTUM, TRIGONELLA FOENUM GRAECUM, CURCUMA LONGA BY ORAL GLUCOSE TOLERANCE AND ORAL STARCH TOLERANCE STUDY IN ALBINO RATS

Abstract: To evaluate the antihyperglycemic effect of Curcuma longa, Ocimum sanctum and Trigonella foenum graecum extracts by oral glucose tolerance and the oral starch tolerance tests on albino rat. Methanol leaf extract of Ocimum sanctum , Curcuma longa rhizomes ethyl acetate extract, methanol seed extract of Trigonella foenum graecum were prepared and supplied by Sami labs, Bangalore . After Institutional animal ethical committee clearance Male albino rats (155–215 g ) were divided into major group A ( Oral glucose tolerance study ) and major group B ( Oral starch tolerance study ). Each major groups consists of 5 sub groups having randomly assigned 6 albino rats . The placebo ( Normal saline ) control group , Standard (Vildagliptin ) group and 3 above mentioned extract groups. After overnight fasting blood glucose level was done (0 minutes). Then placebo, standard and the extracts were administered by using rat oral feeding tube. 30 minutes later oral glucose load (dose 2.2gm/kg ) is administered and sampling was done at 15 and 45minutes interval. Similarly another 5 groups of albino rats were administered with oral starch (3gm/kg ). Blood samples were collected for blood sugar at 0 (before extract ), 30, 60 minutes and blood sugar levels were estimated by enzymatic assay. Trigonella foenum graecum ( 2gm/kg ) shows significant reduction in blood glucose (p= <0.001 ) and Ocimum sanctum (400mg/kg ) ( p= < 0.05 ) at 45 minutes when compared to control. The research extracts shows significant antihyperglycemic effects. Further clinical studies are necessary to establish the therapeutic potential of these extracts in the treatment of type 2 diabetes mellitus. Ocimum sanctum (400mg/kg ) shows reduction in blood glucose ( p= < 0.05 ) at 30 minutes and significant reduction ( p=< 0.001 ) at 45 minutes when compared to Control. Trigonella foenum graecum ( 2gm/kg ) shows significant reduction in blood glucose (p= <0.001 ) at 45 minutes when compared to control. Ocimum sanctum leaf extract, Trigonella foenum graecum seed extract shows significant antihyperglycemic effects. Further clinical studies are necessary to establish the therapeutic potential of these extracts in the treatment of type 2 diabetes mellitus.
