Determination of Suitability of Polymer Matrix for Immobilization of Enzymes

Abstract:

The aim of present study is to establish a suitable polymer matrix for maximum entrapment of enzyme in order to their
safe homing. Carrageenan (1-5%), chitosan (1-5%) and gelatin (5-25%) were chosen for the present study. Beads were
prepared by cross-linking method and enzymes were incorporated in beads by gel entrapment method. Immobilization efficiencies
and working efficiencies of Phospholipase D were found to be 96±3% and 92±1.3% in 3% concentration of chitosan,
55±1.1% and 53±1.9% in 4% concentration of carrageenan and 86±2.3% and 63±2.4% in 10% concentration of gelatin,
respectively. Chitosan was found to be polymer of choice for the immobilization as immobilization efficiency, working
efficiency and physical stability were found to be more as compared to carageenan and gelatin.
