Oral Dispersible Film Containing Eletriptan Hydrobromide for Treatment of Migraine: Design, Formulation, Evaluation and Optimization

Abstract:

The objective of present work was to develop rapidly orally dispersing film of eletriptan hydrobromide for treatment
of migraine headache. The excipients used for preparation of rapidly orally dispersing film were screened for
compatibility with the help of Differential Scanning Calorimetry (DSC) and Oral Dispersible Film (ODF) was
fabricated by solvent casting method using compatible excipients. DSC thermograms do not shown significant change
in endothermic behavior, which indicated all excipients were compatible with the drug.ODF prepared with
Hydroxylpropyl Methylcellulose (HPMC) E-15 was transparent, non-tacky, flexible, disintegrate within specified time,
with good tensile strength. The fabricated film was evaluated for disintegration time, dissolution profile, folding
endurance, tensile strength, surface pH, thickness etc. 32 full factorial design was used for optimization of ODF by
selecting concentration of PG (X1) and concentration of HPMC E-15 (X2) as an independent variables and
disintegration time (Y1) as a dependent variable. The optimized ODF disintegrate within 25.65 sec and around 90%
drug content. Prepared ODF rapidly disintegrate which indicate fast therapeutic effect which required in acute attack
of migraine.
