Abstract:

The aim of this study was to explore the scope of pharmaceutical gum- “Pharmagum® M, to control the drug releases and
to establish the influence of various excipients used in tablet formulations. Valsartan was chosen for incorporation in
matrix tablet and to develop extended release preparation, for treatment of heart failure and post myocardial infarction
patients. The various excipients viz. Microcrystalline cellulose, Colloidal Silicon dioxide, Lactose, Sodium starch glycolate
(SSG) and Silica gel H were tried to check their action on drug release. A 32 full factorial design was selected to optimize
the formulation and the amount of superdisintigrant, Sodium starch glycolate (SSG) (X1) and Silica gel H (X2) were
selected as independent variables. The response (Yi) as percentage drug releases at 4 h and 8 h was measured for each trial
and statistical equations with significant interaction terms were derived to predict Y4 and Y8. Checkpoint batches were
prepared to prove the validity of the evolved mathematical equations. The drug release mechanism was predominantly
found to be Fickian diffusion controlled.
