Suppression the activity of enterotoxin encoding gene Seb in Staphylococcus aureus by Catharanthusroseus extract

Abstract:

Catharanthusroseusis one of the important medicinal plants. It has anti-bacterial, anti-fungal, anti-diabetic, anti-cancer,
due to the presence of indole alkaloids and other biologically active compounds. In this study, we have confirmed that
C.roseushas activity against Staphylococcus aureus, an important pathogen infecting the urinary tract by investigating its
effect on virulence factor expression (enterotoxin). The antibacterial acivity of C.roseus was limited by
Spectrophotometer technique to estimate the MIC through treatment of bacterial culture with different
concentrations of C.roseus extract (10,25,50,75,100 mg/ml) and after incubation period 24 hrs at 37 ?has been
reading the absorbance at 600nm, through the standard curve between the C.roseus extract concentrations and
microbial growth has been shown there are significant differences between the groups which treated with the
concentrations (50,75,100,mg/ml) and (control,10,25 mg/ml) where the microbial growth was more for low
concentrations and less than a high concentrations. A molecular method was used to detect Seb gene which
responsible for enterotoxin production in Staphylococcus aureusisolate by using conventional PCR before and after the
exposure of S. aureusbacteria to the leaves extract of C.roseus at different concentrations (10, 25, 50, 75,100 mg/ml)
through RNA extraction and convert it to cDNA. Results of this study showed a significant effect of C.roseusleaves
extract on Seb gene expression. It has been showed that the expression of Seb gene was decreased at concentration
(75%, 50% mg /ml) respectively at significant level compared to (control, 10%, 25% mg /ml).
