CARDIOPROTECTIVE EFFECTS TO CHRONIC ADMINISTRATION OF GOMPHRENA GLOBOSA FLOWERS IN ISOPROTERENOL INDUCED MYOCARDIAL INFARCTION: BIOCHEMICAL, HISTOPATHOLOGICAL AND ULTRASTRUCTURAL STUDIES

Abstract: The objective of this study is to appraise the cardioprotective properties of Gomphrena globosa flowers on isoproterenol-induced myocardial infarction in albino rats. The experimental rats were divided into four groups with six rats in all the groups. Group 1 and group 2 animals were treated with standard diet for 45 days and will serve as a normal control group and positive control group respectively. Group 3 Rats were administered 2.5 mg/Kg body weight metoprolol succinate by oral route for duration of 45 days. Group 4 Rats received 500 mg/Kg body weight of Gomphrena globosa extract via oral route for duration of 45 days. All the groups except group 1 were then administered 85 mg/kg body weight of isoproterenol by subcutaneous route in two doses on 44th and 45th day at gap of twenty four hrs. At the end of the study period, from all groups blood was collected for the estimation of biochemical parameters by puncturing the retro-orbital plexus and the rats were sacrificed. Serum cardiac marker enzymes for instance creatine kinase (CK), lactate dehydrogenase (LDH), creatine kinase muscle-brain (CK-MB), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total protein, lipid profile which includes high density lipoprotein (HDL), low density lipoprotein (LDL), total cholesterol (TC), triglycerides (TG), and very low density lipoprotein (VLDL) levels were measured. In heart tissue homogenate antioxidant activity was assessed for catalase (CAT), glutathione (GSH), superoxide dismutase (SOD), and malondialdehyde (MDA) levels. Histopathological and ultrastructural studies were then carried out.The present results indicated that ethanolic extract of flowers Gomphrena globosa showed myocardial retrieval by ameliorating the levels of cardiac marker enzymes to normal and increasing the levels of HDL and decreasing the lipid profile. Furthermore, myocardial antioxidants levels were enhanced along with a lessening in the levels of malondialdehyde. The cardioprotective effect was correlated with standard Metoprolol. Following pretreatment with Gomphrena globosa there was reduction in the degree of inflammation and necrosis which was confirmed by histopathological findings. This study concludes that Gomphrena globosa extract exerts cardioprotective activity against isoproterenol-induced cardiac damage in rats
